I love that movie with a deep, abiding love. A couple weeks ago I had some sort of Coen brothers movie virus and watched O Brother Where Art Thou? and The Big Lebowski until my eyes crossed.

"32-20 Blues" is a blues tune by Robert Johnson. It's actually a fairly gruesome song, about a man who plans on killing his unfaithful lover---so it really doesn't have any particular relevance to Sheppard, outside of the obsession with weaponry. (Though Sheppard can be gruesome, when he puts his mind to it.)
